These antigens may be proteins, carbohydrates, glycoproteins, or glycolipids, depending on the blood group system. Some of these antigens are also present on the surface of other types of cells of various tissues. Several of these red blood cell surface antigens can stem from one allele (or an alternative version of a gene) and collectively form a blood group system.  Blood types are inherited and represent contributions from both parents. A total of 35 human blood group systems are now recognized by the International Society of Blood Transfusion (ISBT).  The two most important ones are ABO and the RhD antigen; they determine someone's blood type (A, B, AB and O, with +, − or Null denoting RhD status). As with many other genetic traits, the distribution of ABO and Rh blood groups varies significantly between populations. Transfusion medicine is a specialized branch of hematology that is concerned with the study of blood groups, along with the work of a blood bank to provide a transfusion service for blood and other blood products. Finding your blood type is simple—as long as you have the right test supplies. There are four main blood types: A, B, AB and O, and each of those can be positive or negative for the Rh factor. So in total, there are eight possible blood types: A+ or A-, B+ or B-, AB+ or AB-, and O+ or O-. Materials Required: Monoclonal Antibodies ( Anti-A, B and D) Blood Lancet Alcohol swabs Tooth picks Sterile cotton balls Clean glass slide Ice tray Biohazard disposal container Procedure: Blood Grouping Experiment Set the table with all the materials required. Remember to place the Monoclonal Antibody (Mab) kit in an Ice tray. Open an Alcohol swab, and rub it at the area from where the blood will be sampled (finger tip). (Discard the swab) Open the Lancet cover, put pressure at the tip of the finger from where blood will be sampled (maintain it). Prick the finger tip with the opened Lancet.(Discard the Lancet) As blood starts oozing out, make 1 drop fall on the three depressions of the glass slide. (in clinical setup, there will be a fourth well used as a control). Place a cotton ball at the site where it was pricked. Using the thumb, put pressure on the area to stop blood flow. Take the Anti-A (blue) bottle, resuspend the content and use the dropper to place a drop of the Mab in the 1st spot. Place the bottle back in ice. Take the Anti-B (yellow) bottle, resuspend the content and use the dropper to place a drop of the Mab in the 2nd spot. Place the bottle back in ice. Take the Anti-D (colorless) bottle, resuspend the content and use the dropper to place a drop of the Mab in the 3rd spot. Place the bottle back in ice. Take a tooth pick and mix the content in each well. Discard the tooth pick after using in one well (take a new one for the next well). After mixing, wait for a while to observe the result. Normal Results ABO typing: If your blood cells stick together when mixed with: Anti-A serum, you have type A blood Anti-B serum, you have type B blood Both anti-A and anti-B serums, you have type AB blood If your blood cells do not stick together when anti-A and anti-B are added, you have type O blood. Back typing: If the blood clumps together only when B cells are added to your sample, you have type A blood. If the blood clumps together only when A cells are added to your sample, you have type B blood. If the blood clumps together when either types of cells are added to your sample, you have type O blood. Lack of blood cells sticking together when your sample is mixed with both types of blood indicates you have type AB blood. RH typing: If your blood cells stick together when mixed with anti-Rh serum, you have type Rh-positive blood. If your blood does not clot when mixed with anti-Rh serum, you have type Rh-negative blood. Find a blood type calculator . This video will guide you on how to test blood group at home with the help of Anti A, B and D (Rh) monoclonal antibody reagents. Blood typing procedure: 1. Mix! First mix the each drop of blood with three different reagents including either of the three different antibodies, A, B or Rh antibodies! 2. Look for agglutination! Then you look at what has happened. ... 3. Figure out the ABO blood group! ... 4. Figure out the Rh blood group! ... 5. Figure out the blood type! Blood is often grouped according to the ABO blood typing system. This method breaks blood types down into four categories: Type A Type B Type AB Type O Rh +ve or -ve Blood typing is also done to tell whether or not you have a substance called Rh factor on the surface of your red blood cells. If you have this substance, you are considered Rh+ (positive). Those without it are considered Rh- (negative). Rh typing uses a method similar to ABO typing. Normal Results ABO typing: If your blood cells stick together when mixed with: • Anti-A serum, you have type A blood • Anti-B serum, you have type B blood • Both anti-A and anti-B serums, you have type AB blood If your blood cells do not stick together when anti-A and anti-B are added, you have type O blood. RH typing: • If your blood cells stick together when mixed with anti-Rh serum, you have type Rh-positive blood. • If your blood does not clot when mixed with anti-Rh serum, you have type Rh-negative blood. The last slide blood group is B -ve as you can see clumping/agglutination with Anti-B reagent and no agglutination with rest 2 Considerations There are many antigens besides the major ones (A, B, and Rh). Many minor ones are not routinely detected during blood typing. If they are not detected, you may still have a reaction when receiving certain types of blood, even if the A, B, and Rh antigens are matched. A process called cross-matching followed by a Coombs' test can help detect these minor antigens and is routinely done prior to transfusions, except in emergency situations.

A lancet: which you will use to prick your finger with. A little pump dropper: to drop water on to your Eldon card. Cotton swab: to wipe up any blood Eldon sticks: These are where you will drop your blood from your pricked ear or finger. Alcohol swab Instructions to figure out what our blood type is. How to test your blood type: You put one drop of water on to each one of the colored circles on your Eldon card. Making sure you only put one drop. Gently turn the Lancet cover. Place it on your finger or preferred extremity you will be using to draw blood. Press down till it clicks. Push the lancet in deeper, once you hear the click. Its almost painless. The anticipation is the worst part. Next drop the blood on Eldon Sticks and swish the bloody tip around on each circle on the Eldon Card. Once all the circles are filled wait 10 seconds. Hold and turn the Eldon card for a few second at every square angle. Finally look at the chart and instructions and you will see your blood type according to your blood pattern. Thanks for watching. [UPDATE: I recently used spring-loaded lancets for this lab. They worked great. Here's a video showing my experience with them: https://www.youtube.com/watch?v=v7ps2N0DHG4 This is great to show kids to take away the fear of using this unknown object. :) ] I made this video to show my students my experience with the EldonCard. After I did this experiment with around 75 students, I have some tips that I don't show in this video. 1) If a person is helping you (which I highly recommend) have that person wear gloves. 2) When disinfecting with the alcohol swab, use an outward circular motion (instead of sweeping microorganisms back onto your finger) 3) Use a pipette to get the one drop of water. (Don't use the bottle that Carolina provides) 4) Be properly hydrated before pricking your finger (this allows your blood to flow faster after being pricked) 5) Have everything ready and to do the steps in the correct order instead of by doing by memory (this caused the students' wounds to clot before getting out the blood) 6) The person that is pricked should squeeze out his blood on the EldonStick. The person helping should hold the EldonStick for them while they do this 7) Prick the finger towards the side of the finger, rather than the center. I think this might cause less pain since less nerves are located there. 8) To help get blood out quickly, after being pricked, make sure you are standing up. Place your arm with elbow upwards and hand pointed at the ground (like an upside down "L"). Then with your other hand, make a very tight fist on the pricked finger and squeeze tightly downwards, like you are trying to milk a cow. 9) TILT THE CARD IN THE CORRECT ORDER. The one thing that really messed up some of my students' results is that they didn't tilt upside down first and then right side up. If tilted incorrectly, the water in one field can spill into another field, causing the antibodies from both fields to mix and the results to be inaccurate. 10) Protect your results using clear packing tape if your card doesn't come with "fixation foil" 11) If someone faints at the sight of blood (I had a student faint while watching the instructional video) be sure to allow that person to lay on the ground until they recover. Picking up their legs also helps restore blood flow back to the brain . 12) Using lancets from a glucose meter doesn't work because the prick is way too small to get a decent amount of blood out. Use the lancet that comes with your kit. Not to mention, they give 4 of these bottles for a class of 25 students which makes no sense to me. 2) They provided a DVD for both of the kits I had ordered, which just seemed wasteful. I thought this should be sold separately, since I now have 2 identical DVDs 3) The DVD shows items provided with this kit that aren't provided by Carolina, like a protective cover for your results, a cotton ball, and most importantly, the pipettes! (I'd rather order a kit that comes with the protective cover, a.k.a. fixation foil, for each card; however, I've provided packing tape for my students to tape up their cards which works just as fine I believe.)

These include your digestive tract, from your mouth to your large intestine, as well as your nasal passages and lungs. Because these blood type antigens are everywhere, they influence how your body reacts to the food you eat through several factors. For example: the lectins in certain foods bind to your blood type antigen and cause your blood to agglutinate (stick together), resulting in feelings of fatigue, headaches, digestive issues, skin problems and a host of other health issues. Gut bacteria is related to blood type. Blood typing is done so you can safely donate your blood or receive a blood transfusion. Your blood type is based on whether or not certain proteins are on your red blood cells. These proteins are called antigens. Blood is often grouped according to the ABO blood typing system. The 4 major blood types are: Type A Type B Type AB Type O Test procedure- A blood sample is needed. The test to determine your blood group is called ABO typing. Your blood sample is mixed with antibodies against type A and B blood. Then, the sample is checked to see whether or not the blood cells stick together. If blood cells stick together, it means the blood reacted with one of the antibodies. The second step is called back typing. The liquid part of your blood without cells (serum) is mixed with blood that is known to be type A and type B. People with type A blood have anti-B antibodies. People with type B blood have anti-A antibodies. Type O blood contains both types of antibodies. The 2 steps above can accurately determine your blood type. Rh typing uses a method similar to ABO typing. Blood typing made easy with explanation on ABO blood groups and Rh factor for nurses (blood transfusions). There are 8 total blood types from four blood groups (A, B, AB, O). In nursing we transfuse blood, but before we do this we have to collect blood from the patient who will be receiving the blood transfusion. The patient's blood will be typed and crossmatched with a donor. The donor’s blood must be compatible with our patient to prevent a transfusion reaction. To understand blood typing, you have to understand the relationship between antigens and antibodies. What are red blood cell antigens? They are either present or absent on the surface of RBC. All red blood cells have them EXCEPT Type O RBCs. Antigens are proteins that can elicit an immune response when they come into contact with its corresponding antibodies. Therefore, they stimulate antibodies to defend the body. So, when the same red blood cell antigens and antibodies get together it will cause an IMMUNE RESPONSE called agglutination. This is where the RBCs will glue together, hence clump together. Therefore, it is VERY important a person is not transfused with the wrong blood type. Blood Types: Recipient and Donor A blood type: has only A antigens on its surface with B antibodies in the plasma. Type A: donates to A and AB and recipient of O and A B blood type: has only B antigens on its surface with A antibodies in the plasma. Type B: donates to B and AB and recipient of O and B AB blood type: has both A and B antigens on its surface with NO antibodies in it plasma. Type AB: donates to only other ABs but recipient of O, A, B, and AB...known as the "UNIVERSAL RECIPIENT" O blood type: has NO antigens on its surface with A and B antibodies in its plasma. Type O: donates to all types but only recipient of other O....known as the "UNIVERSAL DONOR". Rh factors: either present or absent on the red blood cells surface. If these factors are present on the RBC the patient is Rh POSTIVIE, but if these factors are absent the patient is Rh NEGATIVE. If a patient is Rh positive they can receive either Rh+ or RH- blood. While Rh negative patients can receive only Rh- blood. So I decided to give my self a home blood test, using the Eldon Home Blood Kit. I have never know what my blood type was so I figured I should find out. Do I have the Rh negative blood type? I've seen a lot of weird and wild theories about the genetic mutation (RH negative blood) online and would be really surprised if I was a rh negative... I always remember my mother telling me I had a very rare blood type and its always fascinated me, the Mystery of the Blood and where the Rhesus Neg came from, as it lacks the Rhesus Monkey Protein and there are some crazy theories regarding it....so we got these home test kits and the results were AMAZING!
